A Phase 1/2 clinical trial has just opened at Moffitt Cancer Center for relapsed/refractory sarcoma and osteosarcoma. This study is evaluating domatinostat, an oral histone deacetylase (HDAC) inhibitor, in combination with sirolimus, an mTOR inhibitor, in adolescents and adults ages 12–40. The Phase 1 dose-escalation portion will determine the maximum tolerated dose and recommended Phase 2 dose in patients with relapsed or refractory sarcoma. The Phase 2 portion will then enroll two osteosarcoma cohorts: patients who have achieved complete surgical remission, and patients with unresectable, measurable disease. The trial is sponsored by H. Lee Moffitt Cancer Center and Research Institute, in collaboration with the National Pediatric Cancer Foundation and Healx USA.
Moffitt Cancer Center in Tampa, FL is currently recruiting. Additional sites — Washington University in St. Louis, Montefiore Medical Center (Bronx, NY), and Nationwide Children's Hospital (Columbus, OH) — are expected to open soon. The study is led by principal investigators Dr. Jonathan Metts at Moffitt Cancer Center and Dr. Amy Armstrong at Washington University School of Medicine.
